OPENED CLINICAL-PHARMACOLOGICAL RESEARCH EFFICACY AND TOLERABILITY
OF ONCE DAILY AMLODIPIN IN TREATMENT ANGINA PECTORIS STABILIS

Objectives:
In this prospectively randomized study we have examined the efficacy,
tolerability, and safety antagonist of calcium channels, which
is containing amlodipine its active component. Methods: After
wash out phase of two weeks 20 patients of an average age 54,529+-8,74
(average +- STDEV), average weight 80 kg and average of duration
of illness 3 years suffering from angina pectoris stabilis participated
in the study. We controlled tolerate of exertion (exercise test
- Bruce protocol), blood pressure, heart rate.We adjusted daily
doses of amlodipine after every clinical axaminations (4 clinical
axaminations in four months), recorded adverse effects of drugs
and obtained specimens for laboratory testing.We didn't approved
using any another antihypertensives and vazodilatators except
oral hlortalidon and nitroglicerol in study period. Results: 17
patients finished study completely.After wash out phase average
number of anginal attacks for all patients was 3,6+-1,8, average
using of nitroglicerol in one week of wash-out phase was 3,8+-1,9,
on last week of clinical examination average number of anginal
attacks for all patinets was 1,4+-0,6, average using of nitroglicerol
was 1,4+-0,63. We recorded good results in average of decreases
blood pressure, heart rate and good tolerate of exercise test
(Bruce protocol). Conclusion: This research has proved good tolerability
of amlodipine with more than 85% of patients as well as its efficacy
in decrease of anginal attacks, tolerate of exercise (Bruce protocol),
lowering blood pressure and based on subjective estimation by
patients, this drug improved life quality of all of them. .
